Mechanism of action
1. Inhibition of Ergosterol Synthesis
Ketoconazole inhibits the fungal enzyme cytochrome
P450-dependent lanosterol 14-a-demethylase (CYP51).
This enzyme converts lanosterol → ergosterol, which is an essential structural component of fungal cell membranes Loss of membrane integrity → impaired fungal growth.
2. Fungistatic and Fungicidal Activity
At lower concentrations: fungistatic (inhibits fungal growth).
At higher concentrations: fungicidal (kills fungal cells).
3. Anti-inflammatory Effect on Skin
